Proteasome analysis presents challenges due to its high molecular weight and low stability. Here, we present a protocol to assess proteasome activity, assembly, and composition in Schizosaccharomyces pombe. We describe steps for lysate preparation and native electrophoresis to separate proteasome complexes.
